Keywords: CHD1, PRO-SEQ, RNA-SEQ, Differential Expression, Dosage Compensation, Transcription Topic of the Thesis: "Study of the Role of the Chromatin Assembly and Remodeling Factor CHD1 in Global Transcription Regulation" Objective of the Thesis: To study the role of CHD1 in global transcription regulation using bioinformatic analysis of RNA sequencing (RNA-Seq) data and transcription sequencing during transcription (PRO-Seq) in a Drosophila model. The following tasks were undertaken to achieve this goal: 1. Investigate the role of CHD1 in the regulation of differential gene expression. 2. Analyze the role of CHD1 in dosage compensation. 3. Examine the role of CHD1 in the regulation of aberrant transcription. 4. Develop a pipeline for analyzing comprehensive transcription regulation using data on nascent transcript sequencing and total transcription. The subjects of the research were S2 cells and larval heads of Drosophila melanogaster. The focus of the study was the global regulation of transcription by the CHD1 factor. The functions of CHD1 were examined using the classical model of dosage compensation and in silico methods for processing PRO-Seq and RNA- Seq data. Reference data were obtained from Flybase and NCBI databases, which were also used for interpreting the results. The results indicate that: 1. CHD1 is involved in differential gene expression, with regulation depending on cell type. 2. The CHD1 protein is stage- and tissue-specifically involved in the regulation of dosage compensation in Drosophila melanogaster. 3. The CHD1 protein moderately influences transcription from cryptic promoters within genes.
